Does this make the image sharper?

No. Converting cannot restore what was already lost. If the image is genuinely too small or too soft, AI Enhance can upscale it with a model, which is a different operation entirely.

Why is the PNG so much bigger?

Because it stores every pixel exactly rather than approximating. That is the whole point of a lossless format, and it is the trade you are making.
