Does the transparent background survive?

Yes. Both formats have a real alpha channel, so transparent and semi-transparent pixels come across exactly as they were. This is the main reason to choose PNG over JPG here.

Will the PNG be much larger?

On a photograph, yes — often several times. On logos, icons and flat graphics, which is what most transparent WebPs are, the difference is small.

Nothing is lost in the PNG encoding, which is lossless. Whatever the WebP already discarded cannot be recovered, but no further quality is given up.
